No. Only Mastercard varient worked for me. The lounge lady tried multiple POS machines to check the offer for Amex but it wasn't available.

Rubyx has 2 lounge per quarter. One on each card Master/AMEX. Might be AMEX network is not enabled.
Did that person try on old POS machine like this one shown below.

UPDATE 03: SEALDAH IRCTC LOUNGE.
In the past one year I have visited Sealdah Longe multiple times. They provide a rail neer bottle , Coffee , and full meal or snacks depending on the time. They provide meal from 12:00 PM. You can choose from CHICKEN, VEG, EGG meal. But if you access the lounge in afternoon then they will provide snacks instead of meal. Overall, my experience has been very good.

I concluded that only Mastercard varient is eligible for lounge twice a quarter. I tried to use Amex in lounge it didn't work. And dremfolk website didn't showed any lounge for amex.
So I believe only mastercard varient is eligible twice a quarter. Anyway I woude be delighted to be proven wrong.

ICICI is lazy to correct their wesbite info.
AMEX probably works in airport lounge. (Haven't confirmed)
Mastercard works in IRCTC and used it twice a quarter. (Confirmed).
If you have plan to visit IRCTC lounge in near future take two cards with you and use amex in the first try to check if it works.

That is what I was planning to do before. Since you confirmed AMEX won't work at railway lounge also agent confirmed it. So now I wanted to take just Mastercard.
